抄録: In order to find a solvent suitable for the hydrodynamic study of glutenin, behaviors of glutenin were observed in several kinds of dispersing solvents. Solvents used were 0.01M acetic acid, 6M dimethylformamide, 6M urea, 3M and 6M guanidine hydrochloride, each containing 0.01M acetic acid. Observations were made through measurements of viscosity, turbidity, difference spectrum, and optical rotatory dispersion. The order of the dispersing ability was 6M guanidine hydrochloride >6M urea >6M dimethylformamide>0.01M acetic acid. As a dispersing medium 6M guanidine hydrochloride was most promising. In 6M urea and 6M dimethylformamide a viscosity change was observed after the solution was made, but concentrated guanidine hydrochloride did not show such a viscosity change. The viscosity change was eliminated by the addition of salt. However, it caused the aggregation of glutenin to some extent. While the inner structure of protein was destroyed in 6M guanidine hydrochloride and 6M urea, the helix content in 6M dimethylformamide was nearly the same as in 0.01M acetic acid.
