An acid-precipitating protein substance was isolated from the culture fluid after growth of B. anthracis in a medium containing no acid-precipitating substances and its in vitro antigenicity, along with some other properties, was examined. 2. The acid-precipitating substance reacted with anti-B. anthracis serum, and antigeni-cally it was quite different from the supernatant fluid obtained from the culture fluid of B. anthracis after removal of the acid-precipitating substance. 3. Relatively concentrated solution of the acid-precipitating substance reacted with anti-B. anthracis serum very strongly, but at the same time reacted weakly with anti-B. megaterium serum. 4. The antigen component, which was connected with the common precipitin reaction observed between the acid-precipitating substance and anti-B. megaterium serum, was different from the one which was connected with the specific precipitin reaction observed between the same substance and anti-B. anthracis serum. 5. The antigenicity of the acid-precipitating substance both to anti-B. anthracis serum and to anti-B. megaterium serum was, to a certain degree, diminished by heat, but not completely destroyed. 6. The antigenicity of the acid-precipitating substance both to anti-B. anthracis serum and to anti-B. megaterium serum was scarcely diminished by digestion with trypsin, but the degree of reaction between the substance and anti-B. anthracis serum was considerably diminished. 7. The acid-precipitating substance was separated into three fractions by ammonium sulfate fractionation and by the adjustment of pH, and the antigenicity of the fractions was examined. The fractions separated showed differences in titers to both anti-B. anthracis and anti-B. megaterium sera. 8. All the fractions showed in their antigenicity more or less a certain degree of influence either by heat or by digestion with trypsin. 9. Of the three fractions, the most active one, referred to as fraction F-3, was compared in antigenicity with the intracellular substance and the capsular polypeptide. The fraction appeared to be related to the intracellular substance, especially to its acid-precipitating component, but it appeared to be not related to the capsular polypeptide. 10. The acid-precipitating substance was positive to the biuret test and electrophoretically it was not homogeneous, while the fraction, F-3, showed one component migrating towards the anode. In the paper chromatographical anslysis six or seven amino acids and two unidentified ninhydrin-positive components were found in the hydrolyzate of the fraction. 11. This study confirms that the acid-precipitating substance separated from the culture fluid after growth of B. anthracis is active as a precipitinogen and that the fractionation of the substance with ammonium sulfate combined with the subsequent adjustment of pH results in the fraction which is more antigenic.
